Director James Gunn said that Chris Pratt's audition was so good he didn't even have to lose weight if he couldn't and they'd just CGI him a six pack if they had to. Ironic, considering Gunn originally didn't even want to see Pratt for the role. Pratt got the audition because Gunn's assistant insisted. Gunn later said that within 30 seconds he knew the role was Pratt's.

It took makeup artists five hours a day to apply the 18 prosthetic tattoo pieces and full body makeup onto Dave Bautista. Chris Pratt said Bautista stood the entire time, holding onto rails with tennis balls in his hands, silent and never complaining. -

Vin Diesel said that his performance as Groot helped him through the difficulty of losing his friend, Paul Walker. Chris Pratt told director James Gunn he'd need six months to lose the 50 pounds for the part. In the end, he actually lost 60 pounds, thanks to a strict diet and a torturous workout regimen. He said it was worth it to see the best possible physical version of himself. -

Vin Diesel recorded all of his "I am Groot" lines in several languages including Russian, Mandarin, Spanish, Portuguese, German, and French so they could use his real voice in every rendition of the film. In all, he ended up recording the line over 1,000 times.

Chris Pratt very proudly likened his role as Star-Lord, and his portrayal of the character, to that of Harrison Ford's Han Solo. Harrison Ford was 35 when he first donned the Solo vest, and Pratt was 35 when he first donned Star-Lord's. -

(#23) James Gunn Gave Out Play-Doh as a Reward
James Gunn kept a pile of Play-Doh canisters on set and would give them out whenever someone did an amazing job that day. Everyone from an actor, a grip, a stunt man, or a PA was eligible. During the 85 day shoot, with a crew of over 200, he gave out just 40 or so Play-Doh rewards.
Gunn said, "I love the smell of Play-Doh, opening a new container and smelling it puts me in a creative, child-like place. Who doesn't love playing with Play-Doh?" -
(#26) James Gunn Spent Lots of Time Fine Tuning the Soundtrack
James Gunn correctly assumed that using iconic and memorable 70s and 80s songs would help ground the film in reality and add a sense of nostalgia. He said he compiled a list of 120 chart topping hits and would even play the songs for the actors on set. Almost every song on the soundtrack was planned to be used even before cameras started to roll. -
(#29) The Guardians Characters Have Been Around For a While
The original Guardians of the Galaxy was comprised of an entirely different team when it debuted in comics in 1969. However, the characters that make up the newer team used in the film were all introduced before the first Star Wars film (to which Guardians is always compared) was released.
Star-Lord and Rocket have both been around since 1976, Gamora since 1975, Drax since 1973, and Groot was introduced all the way back in 1960.

(#32) Where Is Iron Man?
Early drafts of the script featured an Iron Man cameo, which would've tied the film tighter into the current Marvel Cinematic Universe. Plus, every Marvel Studios film does much better with Robert Downey Jr. in it. It's science. On top of that, Iron Man has been a member of the Guardians in the comics.
A number of factors probably went into why Iron Man didn't appear, the film however became the first Marvel Studios film to pass $300 million domestically without RDJ involved.

(#33) James Gunn's Brother Played Rocket on Set
Adam Sandler, David Tennant, Sharlto Copley, Jim Carrey and H. Jon Benjamin were all considered for Rocket Raccoon before Bradley Cooper was cast. Many fans lobbied for Jason Statham since Rocket is described with a similar accent in the comics.
James Gunn's brother, Sean, played Rocket on set during filming, with Cooper's voice added later.

Jason Momoa (Khal Drogo on Game of Thrones) auditioned for, and was then offered the role of Drax the Destroyer, only to turn it down, fearing he would be pigeonholed as only playing brutes. He also wanted more time to focus on directing his first feature, The Road to Poloma. He went on to accept the role of Aquaman for DC Comics. -
(#40) Zoe Saldana Had Already Played A Loser
Peter Quill/Star-Lord refers to their group as "a bunch of losers." Coincidentally, Zoe Saldana had previously starred in a comic book film called The Losers, about a team of heroes. Another member of this team was played by Chris Evans, who plays Marvel's Captain America. -

Djimon Hounsou (who plays Korath) originally auditioned for Drax, and Lee Pace (who plays Ronan the Accuser) originally auditioned for Star-Lord.
