Sure, it's tough to say exactly how cartoon characters would behave in the real world, but in the case of Peter Griffin and Glenn Quagmire, there are some personality quirks that would be hard to overlook. Quagmire's aggressive sexual behavior towards Peter's wife Lois would definitely be a source of contempt - having a friend who's constantly lusting after your wife would certainly cause a couple of buddies to burn bridges.
Maybe they would see one another at an awkward barbecue among mutual friends every few years, but overall, there's no way these two would be routinely going out for beers.

We all have that one friend who loves to push us a little too far, but in the case of Sex and the City's Carrie Bradshaw and Miranda Hobbes, it's hard to believe these two would get to the point where they could even call one another "friend" to begin with.
Miranda is career-focused and cynical, while Carrie can be impulsive, impractical, and narcissistic, which means these two characters probably wouldn't even be in the same social circles.

It's hard to define this one as an actual "friendship," since Dr. Dana Scully was assigned to Agent Fox Mulder's "X-Files" division. But over the years, the two formed a unique relationship after encountering dozens of bizarre paranormal and extraterrestrial occurrences.
Thing is though, they have polar opposite personalities and beliefs, which is enough to squash any friendship in the real world. Mulder is open to virtually any and every type of urban legend, while Scully is steadfast in her convictions that science can prove or disprove any evidence of the supernatural.
They say opposites attract, but these two would repel each other immediately in reality.

(#9) It's Always Sunny in Philadelphia

Schemes upon schemes upon schemes - the gang on It's Always Sunny are more like a dysfunctional family than a functioning group of adult friends. Somehow cosmically tied to one another (probably because no one else in the world would want to be around them), the crew always find ways to back-stab or betray one another, which would certainly end horribly in real life.
Most humans would never poison members of a rival bar, or create a system to be psychologically manipulative towards women, or shove one another into the path of oncoming traffic to try and score World Series tickets - but the gang does. Between the five of them, there's no way they would be actual friends, and chances are one of them would bite the dust due to their wild antics.

(#5) The Good Place

Without diving into any sort of spoiler territory, it's safe to say the four central characters (Eleanor, Chidi, Tahani, and Jason) might not make the best of friends if they ever found themselves out at the same bar in real life. At one point, Eleanor describes each respectively as "[an] Arizona dirt bag, a human turtleneck, a narcissistic monster, and literally the dumbest person I've ever met."
While their chemistry on-screen has undeniably made for some of the best television in years, this is one group whose members wouldn't even be on each others' radars if fate hadn't intervened and thrown them all together.

Comic and television's most dynamic duo would probably not be so close in real life. Although the two share a common bond - both of their parents were tragically killed before their eyes - the grief spiral that drove each of them to become costumed vigilantes probably speaks more to their dangerously unhinged psyches than anything else.
Being Batman and Robin, crime-fighters is one thing- but being Bruce Wayne and Dick Grayson, supremely-wealthy adult man and untrained teenaged mentee with a total of four murdered parents between them, is something else entirely.
